How Do You Prove That a Skincare Product Reduces Pigmentation?

Short answer: A pigmentation claim is substantiated by showing the product measurably affects melanin production or transfer in a relevant skin model — most commonly by measuring melanin release, tyrosinase activity, or melanin content in pigment-producing cells before and after treatment. In-vitro evidence supports mechanistic claims; visible-appearance claims on human skin generally require clinical testing as well.

What gets measured, and why

Melanogenesis pathway markers. Melanin is produced when melanocytes convert tyrosine through a series of enzymatic steps, so tyrosinase activity is a direct upstream measure of pigment production.

Melanin release and content. Measuring how much melanin is produced and transferred to surrounding keratinocytes shows whether an ingredient reduces pigment at the source.

Skin phenotype matters. Pigmentation biology differs meaningfully across skin types, and post-inflammatory hyperpigmentation is both more persistent and more common in melanin-rich skin — so a claim aimed at that audience should be tested in a model that reflects it, not a single generic skin model.

What in-vitro testing cannot tell you

A positive assay result demonstrates biological activity at the cellular level. It does not by itself prove a visible reduction in dark spots on human skin over a consumer-realistic timeframe — that step requires clinical or consumer testing, and claims should be worded to match the evidence actually held.
